Molecular Beacons and Other Hairpin Probes
نویسنده
چکیده
Stem-loop (hairpin) DNA oligomer probes provide increased specificity of target recognition as compared with linear DNA probes. Invention of molecular beacons, a variant of stem-loop oligonucleotides, which are able to fluoresce upon hybridization but are ‘‘dark’’ in the absence of the target, made possible real-time monitoring of hybridization process. A variety of robust assays using diverse, structurally constrained oligonucleotides have been developed that exploit endless number of formats and detection methods. Hairpin DNA oligonucleotides are also used as different components in many DNA amplification methods providing higher specificity and lower background. Thus, these structured probes are an indispensable tool in modern biotechnology and diagnostics.

Exciton interaction in molecular beacons: a sensitive sensor for short range modifications of the nucleic acid structure
Molecular beacons are hairpin-shaped, single-stranded oligonucleotides constituting sensitive fluorescent DNA probes widely used to report the presence of specific nucleic acids. In its closed form the stem of the hairpin holds the fluorophore covalently attached to one end, close to the quencher, which is covalently attached to the other end. Here we report that in the closed form the fluoroph...

Understanding viral pathogenesis is critical for prevention of outbreaks, development of antiviral drugs, and biodefense. Here, we utilize molecular beacons to directly detect the viral genome and characterize a clinical isolate of bovine respiratory syncytial virus (bRSV) in living cells. Molecular beacons are dual-labeled, hairpin oligonucleotide probes with a reporter fluorophore at one end ...
متن کاملThermodynamic basis of the enhanced specificity of structured DNA probes.
Molecular beacons are DNA probes that form a stem-and-loop structure and possess an internally quenched fluorophore. When they bind to complementary nucleic acids, they undergo a conformational transition that switches on their fluorescence. These probes recognize their targets with higher specificity than probes that cannot form a hairpin stem, and they easily discriminate targets that differ ...

The completion of the Humane Genome Project has resulted in an exponential rise in the demand for molecular diagnostic assays. To meet this demand, several innovative technologies have become available for performing homogeneous genetic analyses. For this type of assay, special detector probes are necessary. In 1996, Tyagi and Kramer described fluorogenic hairpin-shaped detector probes, called ...
متن کاملFluorescent hybridization probes for nucleic acid detection.
Due to their high sensitivity and selectivity, minimum interference with living biological systems, and ease of design and synthesis, fluorescent hybridization probes have been widely used to detect nucleic acids both in vivo and in vitro. Molecular beacons (MBs) and binary probes (BPs) are two very important hybridization probes that are designed based on well-established photophysical princip...
